研究方向

1.免疫遗传学

对鱼类粘膜的免疫功能进行探究,采用模式鱼类斑马鱼炎症模型,结合经济鱼类病理分析,针对肠肝轴中菌群、胆汁等对免疫细胞及分子的互动调节,以及脑肠中的神经免疫互动,采用荧光标记品系和整合生物信息学挖掘,并配合无菌斑马鱼和类器官模型,来深入探索粘膜屏障中抵御病原同时构建耐受的免疫调节机理。其中,多组学,包括组织水平转录组的深入分析(如淋巴细胞浸润、细胞通讯等)、单细胞转录组、ceRNA网络等分析可挖掘出关键的调节基因、非编码RNA等,可作为分子育种的靶点。 

2.粘膜免疫学与渔药研发

基于已构建的斑马鱼疾病模型,如肠肝炎症模型,包括IBD(炎症性肠病)和MAFLD(代谢相关脂肪性肝病),采用转录组数据进行中药预测,再将潜在药物添加到疾病模型中,然后采用多组学,包括转录组、菌群组学、代谢组等,并结合流式细胞术分析和表达分析,阐明药理(免疫和代谢机理),并通过系统药理学明确主效成分和靶点基因。同时结合病原性疾病易感性分析,已对多种中草药进行免疫增强剂类渔药的药效和药理分析,解析了应用范围和方式。

科研成果

1.鱼类肠肝炎症模型及免疫的肠肝轴研究

提出了鱼类免疫的肠肝轴,阐明了草鱼食源性肠炎弹性适应的肠肝免疫机理;基于荧光标记免疫细胞成像、基因敲除,建立了斑马鱼肠炎和肝炎模型。

2.渔药研发

基于斑马鱼肠炎模型,通过生信分析获得抗肠炎药物的起效通路,从胆碱能抗炎、肠肝免疫互动等角度,研发了防治肠肝代谢性炎症的药物。对胆碱能抗炎通路,开发了加兰他敏和青藤碱;从肠肝免疫互动角度,从TCMID 数据库中筛选出沙棘。以上成分均已在斑马鱼验证有效并在草鱼中进行了验证及抗菌效果初探,其机理在于通过免疫调节或代谢调节来抗炎,提高了黏膜抗病力。最近,申请人及团队还解析了毛诃子、苦玄参、一枝黄花、紫萁贯众、夏枯草等在抗炎、抗菌、抗寄生虫等方面的机理。

3.粘膜免疫调节机理研究

在免疫调节关键基因突变体中,发现了自发肠炎,获得了黏膜免疫研究的新思路,包括菌群参与的代谢互动、RNA 调节、黏膜免疫与其他器官互动等。从黏膜免疫的代谢调节入手,已揭示SOCS8是抑制肠肝代谢炎症的关键因子;并在免疫的脂代谢调节研究中揭示了没食子酸经由PPARɣ 介导抗炎和调节脂代谢。已揭示的粘膜分子病理还提供了大量关联线索,如代谢物和免疫基因间的关联、肠肝免疫调节的免疫检查点等,为育种提供了分子靶点。

4.已授权/申请发明专利十多项,相关论文发表在Aquaculture、Frontiers in Immunology、Water Biology and Security、Fish and Shellfish Immunology、Frontiers in Physiology、Scientific Reports等;还主持编撰了Frontiers in Immunology的专题并形成了专著。

现任中国水产学会鱼病专业委员会、中国实验动物学会水生实验动物专业委员会、中国细胞生物学会等学术组织的会员,及Frontiers in Immunology编辑;是水产品种创制与高效养殖全国重点实验室、农业农村部淡水养殖病害防治重点实验室的固定成员;兼任宁波大学海洋学院硕士生导师。
